Overview

Triticarte offers a DNA-to-data service: customers provide DNA samples and receive genotypic data.

Confidentiality:
The identity of the customer, identity of the samples and genotypic data are kept confidential at all times. Data are owned by the customer. Triticarte has no IP rights on the data and places no restriction on their use.

Quantity and quality of DNA

For DArT® assays, 500 – 1000 ng of restriction enzyme grade DNA, resuspended in aqueous solution such as TE at a concentration of 50 – 100 ng/microliter should be submitted. As for all genotyping methods, DNA quality has a major effect on DArT® data quality. Therefore, samples are checked for integrity, purity and concentration by running 1 ul of each sample on a 0.8% agarose gel, after incubation in digestion buffer for 1 hour.

Sending DNA samples

The preferred format is 94 samples supplied in sealed 96-well microtiter plates, leaving wells G12 and H12 empty. It is critical that you seal the plate very well. We recommend the use of a standard fully skirted, V-bottom well 96-well plate, sealed with strips of 8 caps, applied carefully one by one and wrapped in glad wrap.

Alternatively, a 3M adhesive film, when properly applied, taking care to seal the wells one by one, should work well. Heat-sealable PCR films DO NOT WORK for this application unless they are heat-sealed properly.

To ensure reliable sample tracking and to protect customer's confidentiality, a sample submission form for automatic input in our Laboratory Information Management System (LIMS) has to be provided by the customer as part of the ordering process.

Australian customers using Australia Post to send samples:
PLEASE DO NOT FORGET to sign the "No Dangerous Good Declaration" on the parcel, otherwise Australia Post will return your shipment to sender.

DArT® data handling

DArTdb, a dedicated database/LIMS, manages the large data sets generated, ensuring data integrity through bar code-based sample-tracking. Our technical and informatics infrastructure is capable of delivering whole genome fingerprints of hundreds of markers for 50,000 DNA samples per year, or about 20 million datapoints. We are in the process of increasing the throughput several-fold through development of further improvements to the technology.

Data reporting
Customers are advised of the completion of the service and download their data from the secure web site. The genotypic data delivered by Triticarte comprise the scores for all markers for each sample analysed. The customer identifies their preferred data format. Most customers currently receive data as an Excel table.

Triticarte will be responsive to customers' needs and welcomes any input on data format of interest to customers.

Additional services (in development)
Triticarte will develop value-adding data processing whereby it will provide various graphical views of the data, build genetic maps, identify marker-trait associations, etc., according to customers' requirements.
